Abstract Bauhinia forficata Link aqueous extract is usually recommended as a phytomedicine to reduce blood glucose levels and its biological activity has been linked to the presence of phenolic compounds from B. forficata preparations. Several drying processes are used in the production of dry herbal extracts, which may influence the chemical composition and efficacy of final herbal medicines. Due to significant chemical changes, defining appropriate drying processes is essential for phytopharmaceutical drug development. In view of this, we analyzed dried B. forficata leaf infusion (BFLI) extracts by HPLC-UV-MSn, followed by molecular networking analysis to evaluate the chemical profiles from dried extracts yielded by freeze-and spray-drying processes. The main metabolites detected included 11 ferulic/isoferulic acid derivatives and 13 glycosylated flavonoids. The qualitative chemical profiles were alike for both drying processes, whereas the relative abundance of some flavonoids was higher using spray-drying. Taken together, our results showed that freeze-and spray-drying preserved the phenolic profile of BFLI and suggested that spray-drying may be the most suitable to obtain its dried products. Along with studying the chemical profiles of dried herbal extracts, evaluating the influence of drying processes on the quality and chemical profiles of final products is pivotal and may benefit future research.

ABSTRACT

O Diabetes desde a antiguidade tem sido uma das maiores causas de morte entre as populações do globo, e segundo a Organização Mundial da Saúde continua assolando nos nossos dias. Apesar das descobertas de tratamentos mais eficazes, a doença vem avançando em progressões assustadoras atualmente, com projeções preocupantes para a saúde pública. Como estratégia de acompanhamento terapêutico, estatístico direcionado a portadores de diabetes, o Governo Federal lançou o programa HIPERDIA (Hipertensos e Diabéticos), que faz o acompanhamento da evolução da doença e das complicações dos pacientes. E neste sentido, também são utilizadas terapêuticas mais acessíveis como as plantas medicinais. O objetivo desta pesquisa consiste em realizar uma revisão bibliográfica abordando as opções de terapias de controle do diabetes oferecidas no Sistema Único de Saúde e pesquisar fitoterápicos com potencial hipoglicêmico aprovados pela Anvisa. Através de levantamento bibliográfico, foram identificadas oito espécies vegetais utilizadas pela medicina popular no controle do diabetes, sendo estas: Bauhinia Forficata, Syzygium Cumini, Annona Muricata, Cynara Scolymus, Momordica Charantia, Eugenia Uniflora e Baccharis Trimera. Essas plantas do programa, embora tenham comprovação de seu efeito hipoglicêmico e redutores dos sintomas diabéticos, pelas suas propriedades antioxidantes e antiinflamatórias, colabora para uma melhor qualidade de vida aos pacientes.


Since antiquity, Diabetes has been one of the biggest causes of death amon-g populations around the globe, and according to the World Health Organization, it continues to plague our days. Despite discoveries of more effective treatments, the disease is currently advancing in frightening progressions, with worrying projections for public health. As a therapeutic, statistical follow-up strategy aimed at people with diabetes, the Federal Government launched the HIPERDIA (Hypertensive and Diabetic) program, which monitors the evolution of the disease and the complications of patients. And in this sense, more accessible therapies such as medicinal plants are also used. The objective of this research is to carry out a literature review addressing the options for diabetes control therapies offered in the Unified Health System and to search for herbal medicines with hypoglycemic potential approved by Anvisa. Through a bibliographical survey, eight plant species used by folk medicine to control diabetes were identified, namely: Bauhinia Forficata, Syzygium Cumini, Annona Muricata, Cynara Scolymus, Momordica Charantia, Eugenia Uniflora and Bacharis Trimera. These plants in the program, although they have evidence of their hypoglycemic effect and reduce diabetic symptoms, due to their antioxidant and anti-inflammatory properties, contribute to a better quality of life for patients.


La diabetes ha sido desde la antigüedad una de las principales causas de muerte entre las poblaciones del planeta, y según la Organización Mundial de la Salud sigue haciendo estragos en nuestros días. A pesar de los descubrimientos de tratamientos más eficaces, la enfermedad avanza actualmente con una progresión aterradora, con proyecciones preocupantes para la salud pública. Como estrategia de seguimiento terapéutico, estadísticamente dirigida a las personas con diabetes, el Gobierno Federal puso en marcha el programa HIPERDIA (Hipertensión y Diabetes), que controla la evolución de la enfermedad y las complicaciones de los pacientes. En este sentido, también se utilizan terapias más accesibles, como las plantas medicinales. El objetivo de esta investigación es realizar una revisión bibliográfica que aborde las opciones de terapias para el control de la diabetes ofrecidas en el Sistema Único de Salud y buscar fitoterapias con potencial hipoglucemiante aprobadas por Anvisa. Mediante un estudio bibliográfico, se identificaron ocho especies vegetales utilizadas por la medicina popular en el control de la diabetes, a saber: Bauhinia Forficata, Syzygium Cumini, Annona Muricata, Cynara Scolymus, Momordica Charantia, Eugenia Uniflora y Baccharis Trimera. Estas plantas del programa, aunque han demostrado su efecto hipoglucemiante y reductor de los síntomas diabéticos, por sus propiedades antioxidantes y antiinflamatorias, colaboran a una mejor calidad de vida para los pacientes.

ABSTRACT

The genus Bauhinia popularly known as "pata-de-vaca", "unha de vaca", "unha de boi", "unha de anta" e "casco de vaca" is widely used in the form of teas and other herbal preparations. The aim of this literature review was to show the diversity and biological potential of Bauhinia species for health promotion. A search was carried out for articles listing some species of medical interest. The pharmacological activities of B. forficata were also highlighted in articles published in the last twenty years using the PubMed database. Research has shown that Bauhinia is used as a hypoglycemic and antidiabetic agent, diuretic, cholesterol reducer, in the treatment of cystitis, intestinal parasites, elephantiasis, tumors and other ailments, including infections and painful processes. In the last eleven years, 86% of the works carried out with B. forficata used the plant collected or acquired in Brazil, predominantly publications from the southern region of the country where almost 60% reported activity on diabetes and its complications and/or antioxidant effect. Despite the literature pointing out the great medicinal potential of Bauhinia in chronic diseases and their complications, there is still a need for more translational research.


O gênero Bauhinia conhecido popularmente como "pata-de-vaca", "unha de vaca", "unha de boi", "unha de anta" e "casco de vaca" é amplamente utilizado em forma de chás e outras preparações fitoterápicas. O objetivo desta revisão de literatura foi mostrar a diversidade e potencial biológico das espécies de Bauhinia para a promoção a saúde. Foi realizada a busca de artigos elencando algumas espécies de interesse médico. Destacou-se também as atividades farmacológicas de B. forficata em artigos publicados nos últimos vinte anos utilizando a base de dados PubMed. A pesquisa mostrou que a Bauhinia é utilizada como hipoglicemiante e antidiabética, diurética, redutora de colesterol, no tratamento da cistite, parasitoses intestinais, elefantíase, tumores e outros males, incluindo infecções e processos dolorosos. Nos últimos onze anos, 86% dos trabalhos realizados com B. forficata utilizaram a planta coletada ou adquirida no Brasil sendo predominante publicações oriundas da região sul do país onde quase 60% relataram atividade sobre o diabetes e suas complicações e/ou efeito antioxidante. Apesar da literatura apontar o grande potencial medicinal da Bauhinia em doenças crônicas e suas complicações ainda há a necessidade de mais pesquisas de caráter translacional.


El género Bauhinia, conocido popularmente como "garra de vaca", "garra de ganado", "garra de tapir" y "pezuña de vaca", se utiliza ampliamente como té y otros preparados fitoterapéuticos. El objetivo de esta revisión bibliográfica era mostrar la diversidad y el potencial biológico de las especies de Bauhinia para la promoción de la salud. Se llevó a cabo una búsqueda de artículos que incluyeran algunas especies de interés médico. Las actividades farmacológicas de B. forficata también fueron destacadas en artículos publicados en los últimos veinte años utilizando la base de datos PubMed. La investigación demostró que la Bauhinia se utiliza como hipoglucemiante y antidiabético, diurético, reductor del colesterol, en el tratamiento de la cistitis, la parasitosis intestinal, la elefantiasis, los tumores y otras dolencias, incluyendo infecciones y procesos dolorosos. En los últimos once años, el 86% de los estudios realizados con B. forficata utilizaron la planta recolectada o adquirida en Brasil, siendo predominantes las publicaciones de la región sur del país, donde casi el 60% reportó actividad sobre la diabetes y sus complicaciones y/o efecto antioxidante. Aunque la bibliografía señala el gran potencial medicinal de la Bauhinia en las enfermedades crónicas y sus complicaciones, todavía es necesario realizar más investigaciones traslacionales.

Bauhinia variegata plant is a very popular and traditionally potent ethnomedicine. Therefore, it is need of hour to study ameliorative characteristics of B. variegata for novel secondary metabolites. The current study was designed to explore antiproliferative potential of B. variegata due to scant reports on this aspect. Extracts of various parts (flowers, leaves, bark, stem, and roots) were prepared by successive maceration using organic solvents in increasing order of polarity (n-hexane, ethyl acetate, methanol, and water). The determination of polyphenolic contents was done by using colorimetric methods while antioxidant potential was measured using reducing power assay. Brine shrimp lethality assay was performed for determining preliminary cytotoxicity and antiproliferative activity against breast cancer MCF-7 cell line using MTT protocols. Moreover, antimicrobial activities were detected by using disc diffusion assay. The alpha-amylase assay was performed to monitor the antidiabetic potential of the plant. In case of phytochemical analysis methanolic extract of leaves and bark showed highest phenolic and flavonoids contents. n-Hexane and ethyl acetate extracts of stem and roots exhibited more than 90% mortality with LD50 ranges between 1-25 µg/mL when studied by brine shrimp lethality assay. n-Hexane and ethyl acetate extracts of roots and stem also showed antiproliferative activity against human breast cancer MCF-7 cell line with IC50 values ranges between 12.10-14.20 µg/mL. Most of the extracts displayed moderately high antibacterial and antifungal activities. The n-hexane extract of roots showed antidiabetic activity with 60.80 ± 0.20% inhibition of alpha-amylase. In sum, these preliminary results will be useful for further compound isolation from selected plant parts for the discovery of antibacterial, antidiabetic, and anticancer lead candidates.


A planta Bauhinia variegata é uma etnomedicina muito popular e tradicionalmente potente. Portanto, as características de melhoria de B. variegata foram estudadas. Foi avaliada a determinação dos teores antioxidantes e polifenólicos. O ensaio de letalidade do camarão de salmoura foi realizado para determinar a citotoxicidade preliminar e a atividade antiproliferativa contra linhas de células de câncer de mama MCF-7 usando protocolos de MTT. Além disso, foram detectadas atividades antimicrobianas. O ensaio da alfa-amilase foi realizado para monitorar o potencial antidiabético da planta. Dentre vinte amostras diferentes, o extrato metanólico (EM) da casca apresentou os maiores teores fenólicos totais. A EM das folhas apresentou excelente conteúdo de flavonoides, atividade antioxidante significativa foi exibida pelo extrato hexânico do caule. O extrato do caule de hexano exibe 77,40% como citotóxico em DL50 10,50 µg/mL quando avaliado através do ensaio de letalidade de artêmia. Extratos de hexano e acetato de etila de raiz e caule mostraram atividade antiproliferativa contra a linhagem celular MCF7 de câncer de mama humano (IC50 12,10-14,20 µg/mL). Para potencial antimicrobiano importante, vários extratos exibiram excelentes atividades antibacteriana e antifúngica, enquanto o extrato de n-hexano da raiz mostrou atividade antidiabética (60,80 ± 0,20% na concentração de 200 µg/mL). Em suma, estes resultados preliminares serão úteis para isolamento adicional de compostos a partir de partes de plantas selecionadas para a descoberta de candidatos a antibacterianos, antidiabéticos e anticâncer.

The pata-de-vaca (Bauhinia divaricata L.) tree is a species widely distributed in Brazil, it has a high ornamental and economic value. Its leaves are used in renal inflammations, such as diuretic, hypoglycemic, being considered a medicinal plant of popular use. The research was carried out in a greenhouse belonging to the Laboratório de Ecologia Vegetal, at the Centro de Ciências Agrárias (CCA), from Universidade Federal of Paraíba (UFPB). The objective was to evaluate the morphophysiological characteristics of B. divaricata plants at different water regimes. The experimental design was completely randomized with five water treatments 1 (100% control), 2 (80%), 3 (60%), 4 (40%) and 5 (20%) of the container capacity. The height, number of leaves and stem diameter were evaluated weekly. At the end of the experiment the plants had their organs separated and taken to the greenhouse to obtain the dry matter of the leaves, stem, roots and total dry matter, and also, biomass allocation in the leaves, stem and roots. Plant height did not differ statistically between the treatments. The number of leaves, diameter, leaf, stem and root allocation and root and shoot dry matter were higher under water availability (100% of container capacity). For the production of B. divaricata L. seedlings water regime should be 100% and 80% of the container capacity, but the seedlings grow satisfactorily.


A pata-de-vaca (Bauhinia divaricata L.) é uma espécie arbórea, amplamente distribuída no Brasil, de alto valor ornamental e econômico. Suas folhas são utilizadas nas inflamações renais, como diuréticas, hipoglicemiantes, sendo considerada uma planta medicinal de uso popular. A pesquisa foi desenvolvida em casa de vegetação pertencente ao Laboratório de Ecologia Vegetal, Centro de Ciências Agrárias (CCA), Universidade Federal da Paraíba (UFPB). O Objetivo foi avaliar as características morfofisiológicas em plantas de Bauhinia divaricata em diferentes regimes hídricos. O delineamento experimental foi inteiramente casualizado com cinco tratamentos hídrico 100%, 80%, 60%, 40% e 20% da capacidade de pote. Avaliou-se, semanalmente a altura, número de folhas e diâmetro do caule. Ao final do experimento as plantas tiveram seus órgãos separados e levados à estufa para obtenção da matéria seca das folhas, caule, raízes e massa seca total, e ainda, alocação de biomassa nas folhas, caule e raízes. A altura de plantas não diferiu estatisticamente entre os tratamentos, o número de folhas, diâmetro, alocação de folhas, caules e raízes e massas seca da raiz e parte aérea foram maiores sob maior disponibilidade de água (100% da capacidade do recipiente). Para a produção de mudas de B. divaricata L. o regime hídrico deve ser de 100% e 80% da capacidade do recipiente, porém as mudas crescem satisfatoriamente.

ABSTRACT Currently, there is a lack of studies on the correct utilization of continuous distributions for dry tropical forests. Therefore, this work aims to investigate the diameter structure of a brazilian tropical dry forest and to select suitable continuous distributions by means of statistic tools for the stand and the main species. Two subsets were randomly selected from 40 plots. Diameter at base height was obtained. The following functions were tested: log-normal; gamma; Weibull 2P and Burr. The best fits were selected by Akaike's information validation criterion. Overall, the diameter distribution of the dry tropical forest was better described by negative exponential curves and positive skewness. The forest studied showed diameter distributions with decreasing probability for larger trees. This behavior was observed for both the main species and the stand. The generalization of the function fitted for the main species show that the development of individual models is needed. The Burr function showed good flexibility to describe the diameter structure of the stand and the behavior of Mimosa ophthalmocentra and Bauhinia cheilantha species. For Poincianella bracteosa, Aspidosperma pyrifolium and Myracrodum urundeuva better fitting was obtained with the log-normal function.

ABSTRACT A new lactone, 7-epi-griffonilide (1), and six known compounds, 2, 3a - 3c, 4a and 4b, were isolated from the leaves of Bauhinia pentandra (Fabaceae). The structures elucidation of 1 and 2 were based on detailed 2D NMR techniques and spectral comparison with related compounds, leading to complete assignment of the 1H and 13C NMR spectra.

ABSTRACT Bauhinia holophylla, commonly known as "cow's hoof", is widely used in Brazilian folk medicine for the diabetes treatment. Therefore, the aim of this study was at evaluating the aqueous extract effect of Bauhinia holophylla leaves treatment on the streptozotocin-induced diabetic rats. Diabetes was induced by Streptozotocin (40 mg/Kg) in female Wistar rats. Oral administration of aqueous extract of Bauhinia holophylla leaves was given to non-diabetic and diabetic rats at a dose of 400 mg/kg during 21 days. On day 17 of treatment, the Oral Glucose Tolerance Test was performed to determine the area under the curve. At the end of the treatment, the animals were anesthetized and blood was collected for serum biochemical parameters analysis. After treatment with Bauhinia holophylla extract, non-diabetic and diabetic rats presented no glycemic changes. On the other hand, the plant treatment decreased body weight and increased ALT and AST activities. In conclusion, the treatment with aqueous extract of B. holophylla leaves given to diabetic rats presented no hypoglycemic effect in nondiabetic animals and no antidiabetic effect in diabetic animals with the doses studied. In addition, the diabetic animals treated with the B. holophylla extract showed inconvenient effects and its indiscriminate consumption requires particular carefulness.

ABSTRACT

Antibiotic resistance results in higher medical costs, prolonged hospital stays and increased mortality and is rising to dangerously high levels in all parts of the world. Therefore, this study aims to search for new antimicrobial agents through bioprospecting of extracts of endophytic fungi from Bauhinia guianensis, a typical Amazonian plant used in combating infections. Seventeen (17) fungi were isolated and as result the methanolic extract of the fungus Exserohilum rostratum showed good activity against the bacteria tested. The polyketide monocerin was isolated by the chromatographic technique, identified by NMR and MS, showing broad antimicrobial spectrum.


La resistencia a los antibióticos conduce a mayores costos médicos, hospitalizaciones prolongadas e incremento de la mortalidad, y está aumentando a niveles peligrosamente altos en todas partes del mundo. Este estudio tuvo como objetivo la búsqueda de nuevos agentes antimicrobianos a través de la bioprospección de extractos de hongos endófitos de Bauhinia guianensis, una planta amazónica típica, utilizada en la lucha contra problemas infecciosos. Fueron aislados 17 hongos; el extracto metanólico del hongo Exserohilum rostratum mostró buena actividad contra las bacterias probadas. Se aisló monocerina policétido por la técnica de cromatografía; este compuesto fue identificado por RM y EM, y mostró un amplio espectro antimicrobiano.

Background: Melanoma is a metastatic type of skin cancer that is difficult to treat and the majority of efforts are directed to the design of new drugs. Medicinal Plants have been the primary source of medicines since life on earth; more than 50% of existing cancer treatments is derived from plants. Bauhinia variegata is well-known medicinal plant used from the ancient era to till date for their medicinal values. Scientific literatures have not documented any evidence of the antitumour potential of Bauhinia variegata against B16F10 melanoma tumor model in C57BL mice. The present investigation was undertaken to explore the antitumour activity of Leaf, stem bark and flower extract of Bauhinia variegata against B16F10 melanoma tumour model in C57BL mice. Methods: Hydro-methanolic extract prepared from the leaf, stem bark and flower of Bauhinia variegata were assessed for their antitumor activity. The extracts at doses of 500 and 750 mg/kg b.wt. were given orally along with cyclophosphamide (chemotherapeutic drug) for 40 days for exploring antitumor activity against melanoma tumor (B16F10) in C57BL mice. Inhibition of tumor growth, increase in survival time of animal with treatment, histopathological studies and antioxidant parameter were determined. Results: The Present investigation showed significant effect of the B. variegata L. in preventing melanoma tumor by B16F10 cell line in C57BL/6 mice. As compared with the tumour control group, the remarkable results especially in the group which received B. variegata extract and cyclophosphamide together were obtained for all of the measured parameters. Dose dependent response was observed in tumor volume, inhibition rate, life span time and antioxidant parameter of extracts. Combination treatment of cyclophosphamide and B. variegata extracts showed more pronounced effect. Conclusions: These findings suggest that B. variegata hydromethanolic extract may contain bioactive compounds of potential therapeutic significance which are relatively safe from toxic effects, and can compromise the medicinal use of this plant in folk medicine (US)

Los hongos endofíticos son hongos que colonizan los tejidos internos de las plantas; varios compuestos biológicamente activos se han aislado a partir de estos hongos. Existen pocos estudios de compuestos aislados de hongos endófitos de plantas amazónicas. Por lo tanto, este estudio tuvo como objetivo el aislamiento y la identificación estructural de ergosterol (1), peróxido de ergosterol (2), mevalonolactona (3), citocalasina B (4) y citocalasina H (5) a partir de Aspergillus spp. EJC 04, un hongo endofítico de Bauhinia guianensis. La citocalasina B (4) y el derivado diacetato de citocalasina B (4a) mostraron una alta letalidad en el ensayo de Artemia salina. Esta es la primera aparición de citocalasinas en hongos endófitos amazónica de B. guianensis


Endophytic fungi are fungi that colonize internal tissues of plants; several biologically active compounds have been isolated from these fungi. There are few studies of compounds isolated from endophytic fungi of Amazon plants. Thus, this study aimed the isolation and structural identification of ergosterol (1), ergosterol peroxide (2), mevalonolactone (3), cytochalasin B (4) and cytochalasin H (5) from Aspergillus sp. EJC 04, an endophytic fungus from Bauhinia guianensis. The cytochalasin B (4) and the diacetate derivative of cytochalasin B (4a) showed high lethality in the brine shrimp assay. This is the first occurrence of cytochalasins in Amazonian endophytic fungi from B. guianensis

Introdução: cepas de Staphylococcus aureus, Escherichia coli e Pseudomonas aeruginosa são caracterizadas como importantes causadores de infeções bacterianas e destacam-se como um preocupante problema de saúde pública. A atividade antibacteriana de várias plantas tem sido estudada com a finalidade de diminuir a resistência microbiana existente com relação aos fármacos já industrializados. O gênero Bauhinia compreende cerca de 300 espécies que são conhecidas no Brasil como pata-de-vaca. A espécie Bauhinia ungulata Lin é bastante utilizada no controle glicêmico, mas, também, apresenta outras atividades, dentre elas é relatado uma ação antibacteriana. Objetivo: verificar o potencial modulador da espécie B. ungulata em associação a outros medicamentos já testados contra cepas de S. aureus, E. coli e P. aeruginosa. Métodos: as folhas da B. ungulata Lin foram coletadas no distrito de Arajara, Chapada do Araripe, estado do Ceará, Brasil. Foi utilizado o extrato aquoso das folhas de Bauhinia ungulata em associação aos antibióticos Amicacina, Gentamicina, Imipenem e Ciprofloxacino, esperando que o extrato agisse com antagonismo ou sinergismo frente ao efeito dos antibióticos. Resultados: o extrato das folhas de B. ungulata Lin apresentou relevante atividade sinérgica sobre os antibióticos utilizados (amicacina, gentamicina, imipenem e ciprofloxacino). Conclusão: portanto, o extrato em associação com os fármacos potencializou o efeito destes, revelando uma atividade antibacteriana significante(AU)

La especie Bauhinia ungulata Lin es ampliamente utilizada en control de la glucemia, pero también ofrece otras actividades, entre ellas es divulgado una acción antibacteriana. Objetivo: verificar el potencial modulador de las especies B. ungulata en asociación con otros fármacos probados contra cepas de S. aureus, E. coli y P. aeruginosa. Métodos: las hojas de B. ungulata Lin fueron recolectados en el distrito de Arajara, Chapada do Araripe, estado de Ceará, Brasil. Se utilizó el extracto acuoso de hojas de B. ungulata en asociación con antibióticos amikacina, gentamicina imipinem y ciprofloxacina, con la esperanza de que el extracto actúa con antagonismo o sinergismo, delantero de los efectos de los antibióticos. Resultados: el extracto de las hojas de B. ungulata Lin presenta relevante actividad sinérgica de los antibióticos utilizados (amikacina, gentamicina, ciprofloxacina y imipinem). Introduction: strains of Staphylococcus aureus, Escherichia coli e Pseudomonas aeruginosa are characterized as important causes of bacterial infections and stand out as a worrying public health problem. The antibacterial activity of several plants has been studied for the purpose of reducing the existing microbial resistance with respect to drugs already industrialized. The genus Bauhinia it comprises about 300 species they are known in Brazil as pata-de-vaca. The specie Bauhinia ungulata Lin is widely used in glycemic control, but also features other activities, among them is reported an antibacterial action. Objective: to verify the potential modulator of the species B. ungulata in association with other drugs tested against strains of S. aureus, E. coli and P. aeruginosa. Methods: The leaves of B. ungulata Lin were collected in the District of Arajara, Chapada do Araripe, Ceará state, Brazil. We used the water extract of leaves of B. ungulata in association with antibiotics amikacin, gentamicin, imipinem and ciprofloxacin, hoping that the extract acted with antagonism or synergism, front of the effects of antibiotics. Results: the extract of the leaves of B. ungulata Lin presented relevant synergistic activity about the antibiotics used (amikacin, gentamicin, ciprofloxacin and imipinem). Conclusion: therefore, the extract in association with the drugs increased the effect of these, revealing a significant antibacterial activity(AU)

ABSTRACT

The genus Bauhinia (Fabaceae, Leguminosae) consists of approximately 300 species, which are commonly known as cows paw or cows hoof, because of the shape of their leaves. They are widely distributed in most tropical countries, including Africa, Asia and South America. Their leaves and stem-bark have been used frequently in folk medicine as a remedy for different kinds of disease, particularly diabetes, infections, pain and inflammatory processes. Bauhinia longifolia flavonoids demonstrated the highest antiviral activity of all tested substances, being that quercetin had the highest antiviral activity amongst purified flavonoids. The aim of this article is to summarize concisely recent advances published in the last 5 years on the most recent works about the biological activities and phytochemical composition of plants of the genusBauhinia. For this, was performed a search in some of the databases on the web as PubMed, Google Scholar and Medline, using the keywords Bauhinia, biological properties, diabetes, cancer(AU)


El género Bauhinia (Fabaceae, Leguminosae) consta de aproximadamente 300 especies, que se conocen comúnmente como pata de vaca o pezuña de vaca, debido a la forma de sus hojas. Están ampliamente distribuidos en la mayoría de los países tropicales, incluyendo África, Asia y América del Sur. Sus hojas y el tallo de corteza se utilizan con frecuencia en la medicina popular como un remedio para diferentes tipos de enfermedades, en particular la diabetes, infecciones, dolor y en los procesos inflamatorios. Los flavonoides de Bauhinia longifolia demostraron la mayor actividad antiviral de todas las sustancias analizadas, y es la quercetina la de mayor actividad antiviral entre los flavonoides purificados. El objetivo de este artículo fue resumir los trabajos publicados en los últimos cinco años sobre la actividad biológica y la composición fitoquímica de las plantas del género Bauhinia. Bauhinia forficata is native to South America and used with relative success in the folk medicine in Brazil. The diversity, antibacterial activity, and extracellular hydrolytic enzymes of endophytic fungi associated with this plant were studied. Plant samples, which included leaves, sepals, stems, and seeds, were used. Ninety-five endophytic fungal were isolated (18 from leaves, 22 from sepals, 46 from stems, and nine from seeds), comprising 28 species. The most frequently isolated species were Acremonium curvulum (9.5%), Aspergillus ochraceus (7.37%), Gibberella fujikuroi (10.53%), Myrothecium verrucaria (10.53%) and Trichoderma piluliferum (7.37%). Diversity and species richness were higher in stem tissues, and Sorensen’s index of similarity between the tissues was low. Eleven fungi showed antibacterial activity. Aspergillus ochraceus, Gibberella baccata, Penicillium commune, and P. glabrum were those with the greatest antibacterial activity against Staphylococcus aureus and/or Streptococcus pyogenes. Thirteen species showed proteolytic activity, particularly Phoma putaminum. Fourteen species were cellulase positive, particularly the Penicillium species and Myrmecridium schulzeri. All isolates tested were xylanase positive and 10 showed lipolytic activity, especially Penicillium glabrum. It is clear that the endophytic fungi from B. forficata have potential for the production of bioactive compounds and may be a source of new therapeutic agents for the effective treatment of diseases in humans, other animals, and plants. To our knowledge, this is the first study of endophytic fungi from different tissues of B. forficata and their biotechnological potential.

ABSTRACT

Metastasis is responsible for the majority of cancer-related deaths. Tumour invasion and metastasis result from processes that include the proteolytic degradation of the extracellular matrix adjacent to the tumour. The matrix metalloproteinases (MMPs), particularly MMP-2 and MMP-9, have prognostic influence in human cancers after they cleave the main structural components of the basal membrane. These actions make MMPs an attractive target for cancer and metastasis studies. This study evaluated the inhibitory potency of extracts of Bauhinia ungulata L. (BU) on the gelatinolytic activity of MMP-2 and -9 and recognized the group of secondary compounds responsible for this property. The zymographic analysis of the BU stem revealed that the ethyl acetate partition (D) caused a higher inhibition of MMP-2 and MMP-9. The phytochemical study of D showed the presence of steroids, tannins, and coumarins and the significant presence of alkaloids and flavonoids. The phytochemical study of the fractions obtained through the column chromatography of partition D revealed a significant presence of flavonoids and alkaloids in the fractions that showed better inhibition of the gelatinolytic activity of MMPs. In conclusion, these results suggest that the stem fraction of BU has the potential to inhibit MMP-2 and MMP-9 and should be used in studies on the recognition of active biomolecules.


A metástase é a responsável pela maioria das mortes relacionadas ao câncer. Tanto a invasão tumoral quanto a metástase resultam de processos que incluem a degradação proteolítica da matriz extracelular adjacente ao tumor. As metaloproteinases de matriz (MMPs), especialmente as MMP-2 e MMP-9, têm influência direta no prognóstico dos diversos tipos de câncer humano, pois clivam os principais componentes estruturais da membrana basal. Estas ações fazem das MMPs alvos atraentes para estudos envolvendo câncer e metástase. Este trabalho teve como objetivo avaliar o potencial inibidor dos extratos de Bauhinia ungulata L. (BU) sobre a atividade gelatinolítica das MMP-2 e 9 e reconhecer o grupo de compostos secundários responsáveis por esta propriedade. Análises por zimograma do ramo de BU revelaram que a partição de acetato de etila (D) causou maior inibição de MMP-2 e MMP-9. O estudo fitoquímico de D mostrou a presença de esteróides, taninos e cumarinas e a presença significativa de alcaloides e flavonoides. As frações da coluna cromatográfica da partição D e seus estudos fitoquímicos revelaram uma grande presença de flavonoides e alcaloides nas frações que apresentaram maior inibição da atividade gelatinolítica de MMPs. Em conclusão, estes resultados sugerem que frações do ramo de BU têm o potencial inibidor de MMP-2 e MMP-9 e devem ser utilizadas em estudos envolvendo o reconhecimento de biomoléculas ativas.

No Brasil, os produtos fitoterápicos são considerados medicamentos, sendo necessário o estabelecimento de estudos que assegurem a manutenção dos requisitos de qualidade durante o processamento e o armazenamento. Testes de dissolução podem ser empregados para se estimar a biodisponibilidade de um fármaco, sendo uma análise rotineira no desenvolvimento e controle de qualidade de medicamentos alopáticos. A determinação do perfil de dissolução de fitoterápicos também pode ser um importante critério para avaliação da sua qualidade lote-a-lote, bem como para os estudos de desenvolvimento e de estabilidade. O objetivo deste trabalho foi investigar a influência dos métodos de secagem e da condição de armazenagem sobre os perfis de dissolução dos flavonoides totais de extratos secos de duas plantas medicinais bastante difundidas no Brasil, a Bauhinia forficata e a Passiflora alata. Os extratos secos foram produzidos pelo processo de secagem em leito de jorro e em spray drying, sendo submetidos a condições de armazenagem aceleradas (temperatura de 40 ± 2ºC e umidade relativa de 75 ± 5%, por um período de 90 dias). Os perfis de dissolução foram obtidos para amostras de extratos secos antes e após o período de armazenamento. O teor de flavonoides totais foi quantificado por espectrofotometria. Os extratos secos de B. forficata e P. alata apresentaram adequada liberação de flavonoides nos ensaios de dissolução. Os extratos secos de Passiflora alata apresentaram completa dissolução dos flavonoides, 92% e 98% dos teores originais após 60 minutos de ensaio, respectivamente para o extrato seco em leito de jorro e em spray drying.


In Brazil, most of the herbal medicinal products are considered as medicine. Therefore, it is necessary the establishment of tests to guarantee the maintenance of quality requirements during their processing and storage. The dissolution test is used to estimate the bioavailability of drugs and is routinely used in the development and the quality control of allopathic medicines. The determination of the dissolution profile of herbal products can also be an important criterion for assessing the batch-to-batch quality as well as for studies of product development and stability. This work aimed to investigate the dissolution profiles of dried extracts of two medicinal plants widely used in Brazil, the Bauhinia forficata and Passiflora alata, by assessing the effect of the drying methods and storage condition on the release of the total flavonoid contents. Spouted bed and spray drying were the processes used for the production of the dried extracts. The products were subjected to accelerated storage conditions (temperature of 40 ± 2ºC and relative humidity of 75 ± 5%, for 90 days). The dissolution profiles of the dried extracts, before and after storage, were determined. The concentration of total flavonoids was quantified by spectrophotometry. Adequate dissolution profiles of flavonoids from B. forficata and P. alata were obtained for all the dried extracts produced. The dried extracts of Passiflora alata showed the complete dissolution of flavonoids in the dissolution media investigated, respectively 92% and 98% of flavonoids present in the dried extracts in spouted bed and spray drying after 60 minutes of the dissolution testing.

Bauhinia forficata Link, popularly known as pata-de- vaca, unha-de-vaca, casco-de-vaca, has been widely used in traditional medicine to treat several diseases. Leaves of B. forficata are used in popular medicine as a diuretic, hypoglycemic, tonic and cleanser, and to combat elephantiasis. However, despite the wide range of ethnopharmacological data surrounding the plant, there are no scientific data demonstrating a probable anti-ulcerogenic activity conferred by use of that species. The present study aimed to evaluate the antiulcer properties of an infusion of fresh leaves of B. forficata Link. From the leaves of B. forficata, an Aqueous extract (AqE) was obtained and the phytochemical analysis showed the presence of flavonols in this extract. In the gastric ulcer induced by administration of HCl- Ethanol model performed with four different doses of AqE (125, 250, 500 and 1000 mg.Kg -1 ), the AqE showed significant preventive activity (*p<0.01) at doses of 1000 mg.Kg -1 . The antiulcer activity of AqE (1000 mg.Kg -1 ) could also be demonstrated in experimental models of NSAID-bethanechol (**p<0.001) and absolute ethanol (**p<0.001). Moreover, AqE (1000 mg.Kg -1 ) promoted a significant increase (**p<0.001) in the amount of gastric mucus. The data presented here demonstrated the potential gastroprotective activity from AqE, possibly attributed to the presence of flavonols in this extract. These results may serve as a s Medicinal plants as an alternative therapy to increase immune resistanceupport for the development of new treatments related to the pathology of gastric ulcer.(AU)

Diabetes mellitus type 2 is a metabolic disorder characterized by chronic hyperglycemia, and an important cardiovascular risk factor with a high morbidity and mortality worldwide. Phytotherapy can be an useful complement in the treatment of chronic diseases, so the hypoglycemic effect of Bauhinia forficata (L) subsp. pruinosa (Fabaceae) was evaluated, as used in Chilean traditional medicine. The content of rutin (R) and trigonelline (T) in aqueous extracts (0.15 and 1.0 percent infusions) and lyophilized extract from leaves of B. forficata were determined by HPLC. It was obtained 2.80 ugR/mL and 2.87 ugT/mL for 0.15 percent infusion, 12.48 ugR/mL and 16.24 ugT/mL for 1.0 percent infusion, and 5.70 ugR/mL and 8.14 ugT/mL for the lyophilized extract (0.1percent). Subsequently, a pilot clinical study in diabetic and prediabetic volunteers (n = 15) was performed, taking one dose of the 0.15 percent infusion, three times a day after meals, for 3 months. The results showed a significant statistically reduction in percent of HbA1c of 0.57 +/- 0.83 (p = 0.0179), suggesting that the 0.15 percent infusion of B. forficata could be useful in the prevention or complementary treatment of diabetes...


La diabetes mellitus tipo 2 es una alteración metabólica caracterizada por hiperglicemia crónica, y un importante factor de riesgo cardiovascular con elevada morbimortalidad a nivel mundial. La Fitoterapia puede ser un complemento útil en el tratamiento de enfermedades crónicas, por lo que se evaluó el efecto hipoglicemiante de Bauhinia forficata (L) subsp. pruinosa (Fabaceae), según su uso en medicina tradicional Chilena. Se determinó mediante HPLC el contenido de rutina (R) y trigonelina (T) en infusos (0.15 por ciento y 1.0 por ciento) y extracto acuoso liofilizado de las hojas de B. forficata. Se obtuvo para el infuso 0.15 por ciento = 2.80 ugR/mL y 2.87 ugT/mL, infuso 1.0 por ciento = 12.48 ugR/mL y 16.24 ugT/mL, y en el extracto liofilizado 0.1 por ciento = 5.70 ugR/mL y 8.14 ugT/mL. Posteriormente, se realizó un estudio clínico piloto en voluntarios diabéticos y prediabéticos (n = 15), tomando una dosis del infuso al 0.15 por ciento, tres veces al día después de las comidas, durante 3 meses. Los resultados mostraron una reducción estadísticamente significativa del por ciento de HbA1c de 0.57 +/- 0.83 (p = 0.0179), sugiriendo que el infuso de hojas de B. forficata (0.15 por ciento) podría ser útil en la prevención o tratamiento complementario de la diabetes...

RESUMO : Este trabalho visou avaliar a ação fitotóxica de extrato e frações obtidas das folhas de Bauhinia ungulata L sobre a divisão celular e atividade das enzimas α-amilase, catalase, peroxidase e polifenoloxidase em plântulas de alface (Lactuca sativa, var Grand rapis). Sementes de alface foram expostas às concentrações de 250, 500 e 1000 µg.mL-1 do extrato de Bauhinia ungulata e frações do mesmo, em condições ambientais controladas. Após três dias de protusão radicular, as raízes das plântulas foram cortadas e submetidas a preparo específico para visualização e contagem do número de células em cada fase mitótica. No sétimo dia, as amostras foram submetidas à avaliação da atividade enzimática utilizando técnica específica para cada enzima e leitura espectrofotométrica. Na análise da divisão celular observou-se que houve redução no número de células em mitose nas raízes das plântulas testadas, e a fração acetato de etila apresentou maior efeito inibitório sobre a divisão celular. O efeito sobre as enzimas que demonstram alterações no sistema de defesa antioxidante de L. sativa mostram que o extrato bruto e frações alteraram a produção das enzimas α- amilase, peroxidase, catalase e polifenol oxidase em, ao menos, uma das concentrações testadas, sendo um indicativo de estresse que interfere na divisão celular. Os resultados obtidos apontam para a presença de compostos com atividade inibidora ou estimulatória nas folhas de B. ungulata revelando potencial fitotóxico sobre as plântulas de alface

Bauhinia ungulata L. (Pata de Vaca) is native to Brazil, mainly found in the Amazon, Cerrado and Atlantic Forest, and popularly used in the treatment of diabetes. This paper describes an analysis of the in vitro antioxidant potential of crude ethanol extract and fractions (hexane, chloroform, ethyl acetate and hydroalcoholic residue) obtained from leaves of this species, using three different methods: 1) scavenging of organic radical 2,2 - diphenyl-1-picrylhydrazyl(DPPH•), 2) reducing phosphomolybdenum complex and 3) assaying the products formed during lipid peroxidation (TBARS). The antioxidant potential of the extract and fractions varied with the methodology, but the ethyl acetate fraction showed activity similar to the standards used in the three methods. A Bauhinia ungulata L. (Pata de Vaca) é uma planta nativa do Brasil, encontrada principalmente na região Amazônica, Cerrado e Mata Atlântica, utilizada popularmente no tratamento do diabetes. Este trabalho descreve uma análise in vitro do potencial antioxidante do extrato bruto etanólico e frações (hexano, clorofórmio, acetato de etila e hidroalcoólica residual) obtidos de folhas desta espécie, através de três metodologias distintas: 1) captura de radical orgânico 2,2-difenil-1-picril-hidrazila-DPPH•, 2) redução do complexo fosfomolibdênio e 3) quantificação de produtos formados durante a peroxidação de lipídios (TBARS). O potencial antioxidante do extrato e frações foi variável de acordo com a metodologia utilizada com destaque para a fração acetato de etila que apresentou atividade similar aos padrões utilizados nos três métodos.
